As loperamide, the most used antidiarrheal drug in Italy, affects the bile ducts physiological motility, it appears interesting to examine the action of ENC on gallbladder smooth muscle motility. The reduction of the gallbladder motility and of the bile flow in patients suffering from pathological conditions that can lead to gallbladder dyskinesia, determine an increase of the risk of cholelitiasis. Otherwise, it is well known that some pathological syndromes, such as cardiovascular diseases correlated to the metabolic syndrome, pregnancy, fibrates therapy strongly predispose to gallstones formation. [2] Drug therapies for the prevention and treatment of gallstones are limited to the use of bile acids. The alternative to drug therapies, in case of symptomatic gallstones, is represented by laparoscopic cholecystectomy. [3] In this work, it has been observed that ENC induces a contraction of the smooth muscle gallbladder in a concentration-dependent manner. In addition, the possible mechanisms involved in the cholecystokinetic activity of ENC have been investigated. ENC gallbladder-mediated contraction has been shown not to be affected by atropine neither by SR27897 suggesting that neither muscarinic receptors nor CCK-1 receptors are not involved in the observed activity. The gallbladder contraction induced by ENC is reduced by nicardipine leading to suppose the involvement of calcium channels in the cholecystokinetic action. In order to value the effects of ENC toward gallbladder smooth muscle motility in pathological conditions, ENC activity has been investigated in gallbladders taken from guinea pigs fed a lithogenic diet. Also in this condition, ENC has been shown to induce the contraction of gallbladder in a concentration-dependent manner. Since there are no drugs able to affect the gallbladder smooth muscle motility, the process leading to the identification of the active compound(s) has been started. References [1] Budriesi, R.
