Strawberry cultivation, for fruit production and related nursery activities, is extensive in the Eastern districts of Emilia Romagna, Northern Italy. Periodic phytopathological problems, despite of the adoption of a certification scheme to produce virus free runners for commercial plantings, are still experienced. Regarding the last three years, some vegetative disorders, as poor plant growth and low fruit production, stunting, decline and other yellows-type strawberry diseases, were observed. Surveys in several fields and nurseries to ascertain the cause of these disorders, as well as the periodical inspection to check the right application of certification rules, were carried out, and samples from plants with or without symptoms were collected. Leaf samples were analysed using biological, serological and molecular methods to identify viral, phytoplasma and other procariote agents. Only two samples out of 202 from certified stocks were positive to Strawberry crinckle virus (SCV), whereas 14 out of 118 (11,9%), collected from fields, resulted infected mainly with Strawberry mild yellow edge virus (SMYEV) and SCV, using biological and molecular tests such as DAS-ELISA, nRT-PCR and Real-Time RT-PCR. Amplification of a specific 16rDNA product from leaf samples with yellows-type symptoms, confirmed the presence of Stolbur (16SrXII-A) in 28 (35%) samples from 78 symptomatic plants collected in nurseries. Furthermore, in 15 samples, coming mainly from strawberry cultivations, the analyses demonstrated the presence of a proteobacterium similar to “Candidatus Phlomobacter fragariae”, agent of the disorder of marginal chlorosis disease, described in France. According to the results from the survey (99%), it seems that plant material with high quality is produced in nursery certified stocks, otherwise material obtained without any sanitary selection show infection range ten-fold higher. Field cultivation are subjected to re-infection by phytoplasma diseases due to vector activities.
