NSAIDs (non-steroid anti-inflammatory drugs) occupy an important position in the classification of the most prescribed and most used drugs in all sectors of clinical practice, and are currently the most used analgesics in Italy, being the first choice drugs for controlling acute moderate pain. Their main clinical application, as anti-inflammatory drugs and pain killers, is for muscular-skeletal disorders, such as rheumatoid arthritis, osteoarthritis and ankylosing spondylitis, headache, trauma and postoperative pain. The main side effect of NSAIDs is at gastric level, including symptoms such as dyspepsia, abdominal pain, mucosal lesions, etc., with gastric bleeding after chronic treatment. Treatment with NSAIDs is highly effective and able to improve the quality of life in a large part of the population affected by acute or chronic disorders; clinical use is, however, often limited due to these potential adverse effects. Topical administration can make it possible to overcome these adverse effects and to maintain plasma levels within the normal range for a long period of time after a single administration. This invention concerns a formulation with an anti-inflammatory activity containing substances commonly used therapeutically, designed to act in synergy in order to achieve a relatively high level of efficacy. The particular formulation provides the topical system with better application properties with respect to traditional formulations, better spreadability and excellent skin-feel. In vitro studies have also shown that the active ingredients are rapidly absorbed while in vivo tests on healthy consenting volunteers have confirmed that the onset of beneficial effects is rapid and effective almost to the same extent as an injectable administration.
