Abstract. Regulated promoters have a pivotal role in synthetic genetic program. To allow the rational design of this part we synthesized a library of operator sequences for LacI, TetR, cI and LexA repressor proteins. Single sequences were cloned as BioBricks (1) to allow their standard assembly with constitutive promoters. Thus, promoter transcriptional strength and repressor binding affinity can be independently fixed. A simple procedure was established to characterize the regulated promoter. These parts are expected to be a benefit in many Synthetic Biology applications.

Abstract. Regulated promoters have a pivotal role in synthetic genetic program.
